The P fraction was evaluated for DPPH radical scavenging activity assays and found to be active with an IC50 43.54 g/mL. The EtOAc fraction showed a strong potent scavenging activity with an IC50 20.75 g/mL in the DPPH ability. The P fraction, however, was more effective than the EtOAc fraction and showed excellent scavenging activity effects of the tap root hairs of a water fern against DPPH free radical.543
We then investigated the antioxidant activities of the active fractions by the DPPH, HOSA, XOS, XOI, NOS, and HPSA assays. The EtOAc fraction was found to be the most effective fraction in scavenging free ^- radical and hydrogen peroxide with IC50 values 18.18 g/mL and 12.12 g/mL, respectively. The IC50 in DPPH assays, XOI assay, NOS assay increased from 19.57 to 42.5 g g, for the P8, after oxidation, respectively. The P8 fraction was also more effective than the EtOAc fraction in preventing oxidation of tap root hair.543
We examined the effects of components of the P8 fraction on the activity of the component of the hair to determine which constituents are more active. The P8 fraction was fractionated into an ethyl acetate fraction (PEA) and an aqueous fraction (PWA). The PWA fraction showed an XOSIC50 value of 1.55 g/mL. The PE fraction also showed strong antioxidant activity such as IC50 19.13 g/mL in the xanthine oxidase inhibition assay and IC50 20.69 g/mL in the nitric oxide scavenging actin.
